The Reds skipper has been absent for the last four games because of a hamstring injury – including a shock 2-0 defeat by Red Star Belgrade last time out and the 1-1 draw with Arsenal at the Emirates.
Henderson, 28, is back in contention for Sunday’s Premier League clash with Fulham at Anfield and could come straight into the side.
And Klopp admits his side are weaker when the midfielder is is not available.
“When Jordan is not available we always miss him,” the Liverpool boss said.
“I would love to say that Hendo would have changed the whole (Red Star) game but I’m not really sure about it.
“The performance altogether over the whole game was strange and not us.”
Despite failing to win any of their last two match, Liverpool are enjoying their best-ever start to a Premier League season – with 27 points from a possible 33.
And Klopp is expecting a reaction against a struggling Fulham side, who are rooted to the bottom of the table.
“We have a wonderful perspective, it’s all good,” he said.
“We had a bad game, we can’t ignore that, but we have the confidence from the games before.
“After Naples we played (Manchester) City, didn’t we?
“That was a good game and the reaction was right.
“We don’t want to lose football games, you always have to show a reaction, that’s clear.
“I’m not in doubt about the reaction. We have 27 points, conceded five goals, done a lot of good things.
“It’s a very positive situation and if we had been in the same situation two years ago, people would have said it’s brilliant.
“It’s no problem that people compare, but for us, it’s important we don’t do it.”
